1. First Purification: The tagged protein binds to beads coated with antibodies that recognize Protein A. After washing away unbound proteins, the TEV protease is used to cut the tag at its specific site, releasing the protein complex.

2. Second Purification: The released complex is then exposed to beads coated with calmodulin, which binds to the CBP part of the tag. After another washing step, the final protein complex is obtained by adding a calcium-chelating agent that disrupts the CBP-calmodulin interaction.
